The only question is what exactly can MR do for Donovan? At this point he's sort of doing dirty things behind the scene and letting Helena stay as the poster child of the newly reformed DOATEC.
Unless in 6, since Helena did find out what Donovan was up to, she has to keep herself heavily guarded and MR was the way into DOATEC to find out what's going on + kill Helena.
What better way to get DOATEC secrets than to be close with the head of it? Relaying information back to Donovan would be easy for Marie Rose because she doesn't look like the typical assassins Donovan sent after Helena. MR is that anime archetype of the cute little girl who is actually evil and has great power. Yeah her damage sucks, but hey, at least she knows how to put up a fight.

Actually during the leon campaign I gave them a reason to make his story of more relevance to the plot (via letter and email)
In short, it turns out that the war Roland dies in is actually because of an army funded by Donovan to open up a secret DOATEC branch in the desert, leon finds out about this due to a Bayman conversation at some point and bam.. he flips out with more rage, and instant story plot.
That can be modified in a hundred ways, but its so easy to plot him into a DOA6 story.
As for Tengu, I did the same, telling them what if he didnt actually die but was banished back into his relm by some ninja magic, and donovan with thirst for different methods of power resummons him.. but this time he is released and uncontrollable and reeking havoc everywhere.
Ein, simply that epsilon actually worked by the end of 5 and that all this time it was actually the prototype clone project. When hayate was freed before completion only the Ein data was extracted.
As for Raidou, ashamed of his defeat in DOA1, he spent years in the forest mastering his skill and is now back for revenge not only on the mugen tenshin, but on all the ninjas.
Genra, could stay dead in a fire. Serves the plot better with a traumatised ayane. His playstyle never fit anyway.

I don't really like the proposal on Tengu and Raidou...
I believe Tengu shouldn't be the same one, but a wholly different one who IS related to the one from DOA2 and being far more powerful than he is and works with Donovan to specifically keep Ryu Hayabusa busy.
As with Raidou, Donovan uses that clone tech to bring him back to adopt his abilities to the Kasumi Alphas as well as use him for data against Ayane and Hayate to see if they are worthy enough for side projects.
Bingo, 2 new villains for all the other ninjas and Rig and Christie can still look credible against the other combatants.

Hmmm, I personally think Marie Rose's story will involve something from the following;
They could do a fair amount with Marie at this point. All we know is she's a servant and she's apparently wicked... which could mean she is literally evil and not one of the good guys, or she just has a devilish personality outside of her job as a servant.... or could just mean she's a practical joker.
I actually hope she ends up working for Donovan. They could do a fair amount with her that way, and could create some interesting relationships.
~ Another possibility is Eliot, who will probably pat her on the head and make a comment about how small she is which will anger Marie to no ends causing a fight to break out... ( Let's face it, this is inevitable. If not Eliot, someone will make a comment about her size causing her to rage. )
- Helena's Servant
- Secretly working for Donovan
- Will have some sort of interaction with Eliot
- Probably has a crush on Donovan's son, Rig.
- Hating on Kokoro somewhere in there

It isn't really fair that Helena's only loyal companion is Zack. She had to outsource Bayman and the ninjas just for extra hands.
Donovan has 6 henchmen.
• Donovan Jr. (Rig)
• His drunk-ass bodyguard (Christie)
• His schizophrenic scientist (Lisa)
• Fame's bitch from Kyoto (Miyako)
• 3rd generation Clonesumi (Phase 4)
• Marie Rose (if ever she was siding with MIST)
And who knows? Kokoro might end up as a receptionist at MIST.

Eh...I don't think Marie is a villain, it's not the fact it doesn't make sense, it's just that if she's a villain, too many people would be villains at this point.
As Raw shows, Donovan still has Kokoro's mother and Lisa(which let's be frank, she isn't dead) on his payroll and has plenty of Kasumi Alpha's AND he has Genra's mask on him which means they have enough powerhouses already and enough of a threat.
Marie could be a deep cover agent, but her intros and outros don't make her as outwardly evil as say Christie is(who doesn't ooze evil, but that feeling is there). I honestly believe she really is Helena's servant who has a sadistic streak.

Excluding outside reinforcements from the ninjas or Bayman, Helena's actually a one woman army at DOATEC. I'm not sure how Zack will play a serious role other than being the chopper pilot.
Would she pick a half-sister that she didn't even know up to this point over her very own mother? Keep in mind that Kokoro was raised by Miyako with a very sheltered upbringing and that she would call out for her mother if things didn't went accordingly her way.
